Stripped-down LSD analogues separate the structural parts that carry the hallucinogenic effect

A team led by David E. Olson at the University of California, Davis synthesised nine simplified analogues by removing structural components from LSD one at a time. Two of them showed profiles with reduced hallucinogenic and cardiotoxic effects in mouse behavioural tests.

On a bright chemistry bench, small clear vials of colourless solution recede out of frame in a metal rack as a blue-gloved hand sets one down, with glass apparatus softly visible behind.

A team synthesised nine simplified analogues by removing structural components

A team led by David E. Olson at the University of California, Davis synthesised nine simplified analogues by removing structural components from LSD one at a time. Olson says taking LSD apart step by step identified simplified cores that are better starting points for medicinal chemistry. The team works at the Institute for Psychedelics and Neurotherapeutics. Bioengineer covered the study, noting that breaking down LSD unlocks new therapeutic discoveries. A team led by David E. UCD0094 and UCD0076 showed profiles with reduced hallucinogenic and cardiotoxic effects

Two of them, UCD0094 and UCD0076, showed profiles with reduced hallucinogenic and cardiotoxic effects. UCD0076 also showed binding skewed towards the 5-HT2C receptor and antipsychotic-like effects in mouse behavioural tests. The peer-reviewed paper appeared in PNAS with DOI 10.1073/pnas.2603412123. The results stay at the level of cells and mice

The results stay at the level of cells and mice and do not amount to a treatment tested in people for schizophrenia, epilepsy or substance use disorder.
