Zipalertinib plus chemotherapy stretched median progression-free survival to 14.5 months

IASLC reports that adding zipalertinib to first-line platinum-pemetrexed raised median progression-free survival in EGFR exon 20 insertion lung cancer from 8.5 months to 14.5 months. PR Newswire's 14 September Seoul dispatch repeats 14.5 versus 8.5 months (HR 0.50). Interim overall survival at 30 percent maturity is HR 0.72 and not yet clear.

14.5 versus 8.5 months

IASLC reports that adding zipalertinib to first-line platinum-pemetrexed chemotherapy raised median progression-free survival in advanced non-small cell lung cancer with EGFR exon 20 insertion mutations from 8.5 months on chemotherapy alone to 14.5 months. The risk of progression or death fell by 50 percent (hazard ratio 0.50). The objective response rate was 65.0 percent versus 40.3 percent. Grade 3 or higher adverse events occurred in 87.1 percent of the combination arm and 54.4 percent of the chemotherapy arm. The results were presented at the World Conference on Lung Cancer in Seoul.[1]

PR Newswire repeats the split and the immature overall survival

A 14 September 2026 PR Newswire report from Seoul says REZILIENT 3 compared zipalertinib 100 mg twice daily plus platinum-pemetrexed with chemotherapy alone. Independent review put median progression-free survival at 14.5 months versus 8.5 months (HR 0.50; P=0.00015), including a brain-metastases subgroup HR of 0.38. Objective response was 65.0 percent versus 40.3 percent, with median duration of response 14.2 versus 9.9 months. Interim overall survival at 30 percent maturity was HR 0.72 (0.42-1.23).[2]

Overall survival is still an interim read

IASLC and PR Newswire agree on 14.5 versus 8.5 months progression-free survival and on HR 0.50. The interim overall-survival HR 0.72 interval 0.42-1.23 does not yet show a clear survival difference. Grade 3 or higher events were more common on the combination in the IASLC report. Neither dispatch treats zipalertinib as a finished overall-survival win.[1], [2]
